Research on reactive changes in the ovaries in response to electron irradiation and testing of radioprotective agents, such as platelet-rich plasma, remains relevant.</p> <p><bold>AIM: </bold>This study aimed to assess the inflammatory response in the ovary after platelet-rich plasma administration in a radiation-induced ovarian failure model.</p> <p><bold>MATERIALS AND METHODS:</bold> Fragments of the ovaries of four groups (I, control (n=10); II, fractional irradiation with electrons in a total dose of 20 Gy (n=10); III, fractional irradiation with electrons in a total dose of 20 Gy + platelet-rich plasma (n=10), and IV, platelet-rich plasma (n=10)) were studied histologically and immunohistochemically using antibodies to pro- (interleukin [IL]-1 and IL-6) and anti-inflammatory (IL-4, IL-10) cytokines, as well as CD3 and CD20.</p> <p><bold>RESULTS: </bold>The immunohistochemical study revealed that electron irradiation led to an increase in the expression of both pro- and anti-inflammatory cytokines and the number of CD3<sup>+</sup> and CD20<sup>+</sup> immunocompetent cells in the interstitial tissue of the ovaries, fractionally irradiated with electrons at a total dose of 20 Gy. The T-cell component was predominant over the B-cell component. Moreover, pre-irradiation administration of platelet-rich plasma contributed to a smaller change in the degree of morphological changes, expression of pro- (IL-1 and IL-6) and anti-inflammatory (IL-4 and IL-10) cytokines, and proportion of CD3<sup>+</sup> and CD20<sup>+</sup> immunocompetent cells in the interstitial tissue of the ovaries. In addition, the T-cell component of immunity was predominant.</p> <p><bold>CONCLUSION: </bold>Components of platelet-rich plasma, having anti-inflammatory and radioprotective properties, reduce the severity of inflammatory response (based on expression levels of pro- and anti-inflammatory cytokines) and number of T and Bimmunocompetent cells, which slow down the development of radiation-induced ovarian failure when exposed to fractional local irradiation with electrons in a total dose of 20 Gy.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Обоснование. </bold>Радиотерапия злокачественных новообразований органов малого таза может приводить к радиационно-индуцированному повреждению здоровых тканей яичника с развитием преждевременной недостаточности яичников и бесплодия. Актуальными остаются исследования, посвящённые оценке реактивных изменений в яичниках в ответ на облучение электронами, а также апробация радиопротекторных средств, одним из которых является плазма, обогащённая тромбоцитами.</p> <p><bold>Цель исследования </bold>— иммуногистохимический анализ воспалительной реакции в яичнике после введения плазмы, обогащённой тромбоцитами, в модели радиационно-индуцированной овариальной недостаточности.</p> <p><bold>Материалы и методы. </bold>С использованием гистологического метода и иммуногистохимически с антителами к про- (IL-1, IL-6) и противовоспалительным (IL-4, IL-10) цитокинам, а также CD3 и CD20 исследовали фрагменты яичников самок крыс породы Вистар в возрасте 9–10 нед. Сформированы четыре группы: I — контроль (<italic>n</italic>=10); II — фракционное облучение электронами в суммарной дозе 20 Гр (<italic>n</italic>=10); III — фракционное облучение электронами в суммарной дозе 20 Гр + плазма, обогащённая тромбоцитами (<italic>n</italic>=10); IV — плазма, обогащённая тромбоцитами (<italic>n</italic>=10).</p> <p><bold>Результаты. </bold>По результатам иммуногистохимического исследования установлено, что облучение электронами привело к увеличению экспрессии как про-, так и противовоспалительных цитокинов, а также к увеличению числа CD3<sup>+</sup> и CD20<sup>+</sup> иммунокомпетентных клеток в интерстициальной ткани яичников, фракционно облучённых электронами в суммарной дозе 20 Гр. Наблюдали преобладание Т-клеточного звена иммунитета над В-клеточным. В то же время предлучевое введение плазмы, обогащённой тромбоцитами, снижало степень морфологических изменений, экспрессии про- (IL-1, IL-6) и противовоспалительных (IL-4, IL-10) цитокинов, а также доли CD3<sup>+</sup> и CD20<sup>+</sup> иммунокомпетентных клеток в интерстициальной ткани яичников. При этом отмечали и преобладание Т-клеточного звена иммунитета.</p> <p><bold>Заключение. </bold>Компоненты плазмы, обогащённой тромбоцитами, обладая противовоспалительными и радиопротекторными свойствами, уменьшают выраженность воспалительной реакции (на основании анализа уровней экспрессии про- и противовоспалительных цитокинов) и число Т- и В-иммунокомпетентных клеток, что приводит к замедлению развития радиационно-индуцированной овариальной недостаточности при воздействии фракционного локального облучения электронами в суммарной дозе 20 Гр.</p></trans-abstract><trans-abstract xml:lang="zh"><p/></trans-abstract><kwd-group xml:lang="en"><kwd>ovary</kwd><kwd>radiation-induced ovarian failure</kwd><kwd>electrons</kwd><kwd>inflammation</kwd><kwd>interleukins</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>яичник крысы</kwd><kwd>радиационно-индуцированная овариальная недостаточность</kwd><kwd>электроны</kwd><kwd>воспаление</kwd><kwd>интерлейкины</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><citation-alternatives><mixed-citation xml:lang="en">practice of radiation oncology: a narrative review.
