Telocyte-like cells have attracted particular attention due to their multifunctionality. Immunohistochemistry is one of the methods for their visualization. Notably, their immunophenotype varies not only in different organs but also in different areas of the same organ. Despite the existing problems with the search for telocyte-specific antigens, double immunolabeling with CD117 and CD34 antigens allows us to consider CD34/CD117 immunopositive cells as telocyte-like with a high degree of probability. Study Objective. To identify and characterize telocyte-like cells at the stages of mechanical skin wound healing in an experiment using transmission electron microscopy and immunofluorescence. Methods. An experimental single-center, continuous, controlled, randomized, unblinded study was conducted. Material: thigh skin of Wistar rats at the stages of incised wound healing. The animals were divided into 9 groups: the control group consisted of intact rats (n=5); other groups corresponded to the experimental periods — 12 h, 24 h, 2, 3, 6, 10, 15 and 25 days (n=5 in each group). Skin fragments were used to prepare preparations for electron microscopy and immunofluorescence studies. Telocyte-like cells were identified in electron diffraction patterns and the expression of two antibodies (CD34, CD117) was studied with double immunofluorescence of the identified cells. The images were processed in Zen 2.3 software for morphometric characteristics and calculation of the expression levels of the markers used. Results. CD34/CD117 immunopositive cells, telocytes, were found in the dermis of rats. This is a heterogeneous cell population with different morphological characteristics. They have different localization, spatial orientation characteristics and express CD34 and CD117 with different intensity at different stages of the wound process. Peak expression of the CD117 marker occurs at 12 hours and 3 days post-injury, while peak expression of CD34 occurs at 15 days post-injury. Conclusion. The study results indicate reactive changes in telocyte-like cells of skin connective tissues during the stages of regenerative prohistogenesis and histogenesis. This indicates the need for further study of the cytophysiological characteristics of this cellular differentiation during wound healing. The most appropriate methods for visualizing telocyte-like cells in tissues include immunohistochemistry, transmission electron microscopy, and immunofluorescence microscopy.</title>
<trans-abstract xml:lang="ru"><p/><title>Обоснование. Наиболее адекватными методами визуализации телоцитоподобных клеток в тканях являются иммуногистохимия, трансмиссионная электронная и иммунофлуоресцентная микроскопия.</title></trans-abstract><trans-abstract xml:lang="zh"><p/></trans-abstract><kwd-group xml:lang="en"><kwd>Skin</kwd><kwd>dermis</kwd><kwd>hypodermis</kwd><kwd>telocytes</kwd><kwd>wound healing</kwd><kwd>immunohistochemistry</kwd><kwd>immunofluorescence</kwd><kwd>CD34</kwd><kwd>CD117</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>Кожа</kwd><kwd>дерма</kwd><kwd>гиподермис</kwd><kwd>телоциты</kwd><kwd>раневой процесс</kwd><kwd>иммуногистохимия</kwd><kwd>иммунофлуоресценция</kwd><kwd>CD34</kwd><kwd>CD117.</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Borges LF, Falcão RSP, Taboga SR et al.
