CHARACTERISTIC OF SEROTONINERGIC NEURONS OF MEDULLARY NUCLEUS RAPHE OBSCURUS IN NORM AND IN SEROTONINERGIC SYSTEM DEFICIENCY DURING THE PRENATAL PERIOD OF DEVELOPMENT IN RATS

Khozhai L.I.

Abstract

Morphological characteristics of the serotoninergic neurons forming nucleus raphe obscurus (NRO), were studied in rats at the early stages (days 5, 10, 12 and 14) of the postnatal period in normal rats and in animals whose prenatal development took place under the conditions of serotonin deficiency. NRO was found to contain three subpopulations serotonin-producing neurons (large, medium and small), which had different sensitivity to serotonin level during development. The results have shown that serotoninergic system deficiency during the prenatal period resulted in the changes of NRO structural organization and in the decrease of the rate of this nucleus formation, serotonin-producing neurons differentiation and the reduction of their total number by approximately a factor of 1.6. At the same time, the significant changes of the dimensions of serotoninergic neurons of all types took place. In control animals, the size of large, medium and small neurons was 1.8, 1.4 and 1.5 times greater than that in experimental animals, respectively. Reduction of the neuron dimensions was associated with the changes of a nucleo-cytoplasmic ratio. The volume of the cytoplasm and of Nissl bodies was significantly decreased. Along with it, the cell destruction was noted that increased with age. Synchronously with it, the marked astrocytic reaction developed, which could further lead to gliosis.

STRUCTURAL AND FUNCTIONAL PECULIARITIES OF THE OF INGUINAL LYMPH NODES AND LYMPH FLOW DURING AGING

Gorchakova O.V., Gorchakov V.N.

Abstract

The comparative experimental study of the lymph drainage and the structural organization of the inguinal lymph nodes (LN) was performed in 160 Wistar rats aged 3–5 and 12–15 months. Age-related changes in inguinal LN were demonstrated that were associated with the reduction of the medium LN and the directivity of lymph flow to the polar LN of the inguinal group. The aged animals showed the decelerated lymph flow and lymph passage through LN due to age-related LN transformation that included the development of the connective tissue, various degree of of structural and functional zone compaction, rarefication of the paracortical zone structure and reduced of lymphocytopoiesis in germinal centers. These findings indicate the development of a state of functional strain of LN with age.

THE PECULIARITIES OF THE THYMUS AND CRANIAL MESENTERIC LYMPH NODE STRUCTURE IN THE NEWBORN RATS AFTER PRENATAL EXPOSURE TO ETHANOL

Pugach P.V., Kruglov S.V., Karelina N.R.

Abstract

The structural peculiarities of the thymus and cranial mesenteric lymph nodes (LN) were examined in 72 neonatal rats born to females, exposed to 15% ethanol solution during pregnancy (n=34), as well as both during pregnancy and for 1 month preceding it (n=38). 44 neonatal rats born to intact females, served as a control group. It was shown that the exposure of female rats to ethanol only during pregnancy resulted in more pronounced changes in the mesenteric LN of the offspring than in the thymus. These included the reduction of LN number, cross-sectional area, lymphoid cell relative content, the suppression of mitotic activity, increased reticular cell content and macrophage activity. Pre-gravid ethanol intoxication of females for 1 month in combination with ethanol exposure during pregnancy was shown to have greater effect on the thymus morphogenesis in the offspring. This was manifested by the decrease of its absolute and relative mass, proliferative activity, increase of cell death, appearance of the microcirculatory and dysplastic changes.

MORPHO-FUNCTIONAL CHARACTERISTIC OF THE DUODENAL CRYPT EPITHELIUM IN THE OFFSPRING OF FEMALE RATS WITH EXPERIMENTAL LIVER INJURY

Serysheva O.Y., Briukhin G.V.

Abstract

The quantitative characteristics and qualitative composition of the crypt epithelium of the duodenal mucosa was studied in 90 Wistar rats aged 1–60 days which were born to females with experimental D-galactosamine-induced liver injury. Histological sections were stained with hematoxylin–eosin, toluidine blue to demonstrate the goblet cells and impregnated with silver nitrate to identify the endocrine cells. The linear dimensions of the intestinal crypts (height and width of the crypts in their longitudinal sections) were determined. The total number of epithelial cells, microvillous enterocytes, endocrine goblet cells, and intraepithelial lymphocytes were counted per longitudinal section through the crypt. Linear dimensions of microvillous enterocytes (height, width), and the volume of their nuclei were calculated. In experimental animals, as compared to the control ones (the offspring of intact mothers; n=90), the linear dimensions of the crypts were unchanged. These animals had the increased number of the microvillous enterocytes, goblet and endocrine cells along with the decreased dimensions of the the microvillous enterocytes and their nuclear volume. At the same time, these animals demonstrated an increase in the nuclear hyperchromism of the epithelial cells and the increment of the intraepithelial lymphocyte numbers.

HEPATIC STRUCTURE REMODELING AFTER AN EXPERIMENTAL PULMONARY TRUNK STENOSIS AND FOLLOWING ITS OPERATIVE CORRECTION

Shormanov S.V., Kulikov S.V.

Abstract

Changes in the liver were studied in 25 puppies with experimental pulmonary trunk stenosis of 6–12 months duration and 10 animals after the elimination of this defect. Control group included 10 dogs of the corresponding age. A complex of histological, morphometric, electron microscopic and immunohistochemical methods was used. During the modeling of pulmonary trunk stenosis, the resistance of hepatic afferent vessels to hepatic blood flow was increased due to the venous-arterial and venous-venous reactions. In the arteries, the bundles of smooth myocytes (SM) of the intimal muscle were formed together with the musculo-elastic sphincters, polypoid cushions, while in the branches of the portal vein, the intimal SM bundles and the valves appeared. In the efferent veins, the muscular elevations were hypertrophied. In all the vessels the thickening of the walls was observed, and in the media of the arteries, there were signs of sclerosis and the increased expression of α-smooth muscle actin (α-SMA). Hepatocytes demonstrated marked ultrastructural changes: mitochondrial matrix swelling, partial destructions of their cristae, dilation of endoplasmic reticulum cisterns. After the elimination of the defect, previously formed vascular adaptation reactions were found to disappear, the tone of the blod vessels in the liver decreased, causing the regression of hypertrophic changes of their media. The number of the arterial blood vessels with intimal muscle, sphincters and cushions decreased. The expression of α-SMA in the media of the arteries was also reduced. In hepatic efferent veins, the muscular elevations became attenuated. The dystrophic changes in hepatocytes regressed at both light-microscopic and the ultrastructural level.

COPPER NANOPARTICLES AS MODULATORS OF APOPTOSIS AND STRUCTURAL CHANGES IN SOME ORGANS

Sizova Y.A., Miroshnikov S.A., Polyakova V.S., Lebedev S.V., Glushchenko N.N.

Abstract

The effect of repeated intramuscular injection into the organism of copper nanoparticles (CNP) with the diameter of 103 nm on the index of cell readiness to apoptosis and the structure of liver, spleen, kidney, as well as sensomotor cerebral cortex, was studied in 78 male Wistar rats. CNP were injected once per week for 12 weeks. The organs were studied using histological, immunohistochemical and morphometric methods. It was found that after the injections, CNP were distributed into organs and tissues of animals causing structural changes that were specific for eaach tissue. Toxicity of CNP in respect to microgliocytes was demonstrated at a dose of 2 mg/kg, hepatotoxicity and nephrotoxicity — at 6 mg/kg. The increase of CNP load on the organism up to toxic threshold (maximum tolerated dose) resulted in the appearance of signs of dystrophy and tissue necrosis. The data obtained suggest the application of an index of cell readiness to apoptosis, as assessed by caspase 3 expression, as a criterion for evaluation of CNP injection safety.

PECULIARITIES OF TISSUE REACTIONS DURING RESORPTION OF BIODEGRADABLE SUTURE MATERIALS

Kuznetsova I.V., Maiborodin I.V., Shevela A.I., Barannik M.I., Manayev A.A., Brombin A.I., Maiborodina V.I.

Abstract

The reaction of the surrounding tissues to the implantation of biodegradable suture materials (plain catgut threads, DemeTECH polyglactin 910, Surgilactin — polyglactin 910) into the subcutaneous adipose tissue of rats, was studied using light microscopy 1, 2, 6 and 12 months after grafting. It was shown that the tissue reactions to implantation of different biodegradable suture materials followed the general pattern characteristic of wound healing and foreign body reaction. By the first month of observation, in all the animal groups, the stage of traumatic perifocal inflammation was replaced by a proliferative phase with the new growth of the blood vessels and connective tissue. In all the animal groups, there was a trend to reduction of productive inflammation activity from the 1 st month till the 12 th month. By the 12 th month the total degeneration of the material with full recovery of structure of surrounding tissues (restitution) was recorded only after the implantation of polyglactin 910 (DemeTECH).

COMPARATIVE CHARACTERISTIC OF MORPHOMETRIC PARAMETERS OF THE ORBITAL OPENING IN MEN AND WOMEN

Gaivoronskiy I.V., Kirillova M.P.

Abstract

100 skulls of adult persons of both sexes were studied of in order to develop a comprehensive morphometric evaluation of the orbital opening (OO) and make a comparative analysis of its parameters in men and women. Сranioscopic, craniometric methods and digital photogrammetry were applied. The data on the sizes of OO and its spatial organization were collected. It was shown that the absolute values of OO sizes were larger in male samples, while its proportions were similar in men and women. Sex differences in OO were demonstrated, in particular, in the parameter of orbital openness. In men, higher values of OO slope were demonstrated, while women had larger orbital openness. Based on this research, the forms of OO and of interorbital space were distinguished, and the categories of angle values of spatial organization were defined.

MORPHOLOGICAL CHANGES OF THE INTERVERTEBRAL DISKS OF THE SPINE CAUDAL PORTION IN RATS AFTER PERMANENT AND TEMPORARY ASYMMETRIC COMPRESSION-DISTENSION

Grigorovskiy V.V., Khizhnyak M.V., Gafiychuk Y.G.

Abstract

In the experiment on 44 rats, histological peculiarities of tissue injury and repair were studied in the segments of caudal spine after long-term permanent and temporary (with the subsequent tale relaxation) asymmetric compression-distension (ASCD). Permanent ASCD of the tail was found to result in a complex of structural changes that were rather stable and did not disappear completely after ASCD discontinuation and a long-term relaxation. These changes included necrosis of the annulus fibrosus (AF) cells, tension of AF lamellae and their separation from epiphyses on the distension side, crushing and splitting of AF collagen lamellae on the side of compression and chondrocyte proliferation around necrotic foci, increase of necrotic notochordal cells number in nucleus pulposus (NP). Relaxation after temporary ASCD resulted in the reduction but not total disappearance of the signs of injury. Changes were noted in AF thickness, occurrence of chondrocyte necroses and the sites of collagen lamellae separation, as well as in NP displacement to distension side.

PECULIARITIES OF DIGITAL DERMATOGLYPHICS AND THE SOMATIC STATUS IN FEMALE ATHLETES ENGAGED IN FENCING

Valkovich E.I., Oleinik Y.A.

Abstract

The article presents the morphogenetic performance of 31 female athlete aged 18–23 years specializing in fencing, compared to women of the same age group not involved in professional sports. The research program included: evaluation of a complex of anthropometric parameters (longitudinal, transverse, circumferential body sizes), somatotype diagnostics using of Heath–Carter method, evaluation of body composition, qualitative and quantitative characteristics of digital dermatoglyphics (pattern type, ridge count, delta index, a combination of phenotypic patterns), determination of sex chromatin content in the epithelial cells of the oral cavity mucous membrane. The study demonstrated that the somatic status of female fencers could be defined as a balanced mesomorphic somatotype with the prevailing mesomorphic vector. The proportion of muscular component in female athletes was higher, while that of the fat mass was lower than the similar parameters in the control group. The athletes were characterized by the peculiarities of dermatoglyphic constitution: high values of delta index and the total ridge count, higher proportion of complex patterns and minimal amount of simple patterns. In female athletes, significantly lower amounts of sex chromatin were demonstrated as compared to those not engaged in sports.

DECALCIFICATION IN FORMIC ACID AFTER FIXATION IN ZINC-ETHANOLFORMALDEHYDE DOES NOT PRECLUDE IDENTIFICATION OF NEURONAL AND GLIAL MARKERS

Kolos E.A., Korzhevskiy D.E.

Abstract

The aim of this study was to determine the possibility of immunohistochemical demonstration of various tissue antigens in paraffin sections of zinc-ethanol-formaldehyde-fixed spinal cord, surrounded by the vertebral tissues and adjacent muscles, after decalcification in formic acid, in adult (n=5) and newborn (n=2) Wistar rats. In the present work, the antibodies used were against: glial fibrillary acidic protein, neurofilaments, neuronal specific nuclear protein, enzyme tyrosine hydroxylase, protein of synaptic vesicles — synaptophysin and the intermediate filament protein — vimentin. Combination of fixation in zinc-ethanolformaldehyde and decalcification in 21% formic acid solution was shown to provide an optimal preservation of the antigens studied.

BONE MINERAL DENSITY AS A SIGN OF AGERELATED INVOLUTION OF HUMAN BONES

Yurchenko M.A., Zolotenkova G.V., Pigolkin Y.I., Fedulova M.V.

Abstract

This review of the literature analyzes the current state of a problem of application of various morphological methods for the investigation of age-related involution of human bone system and their use in the course of forensic personality identification. The difficulties of the application of the available methods of quantitative morphology in determination of biological age, are emphasized. The possibilities of the practical use of radiological methods to study the skeleton, including the determination of the bone mineral density as an indicator of age-related changes of bone tissue, are analyzed. An ambiguity of the research results obtained is noted, and the possible reasons for and the factors contributing to the results variability are considered. It is concluded that the application of computer tomogram densitometry to the various regions of the bone skeleton in complex with some other methods, is promising for the determination of age interval in forensic identification examinations.

POSTNATAL NEUROGENESIS: CELLS IDENTIFICATION AND NOMENCLATURE

Korzhevskiy D.E., Kirik O.V., Gilerovich Y.G.

Abstract

Elaboration of the concept of neural stem cells at the end of the twentieth century resulted in the appearance of the multitude of new terms used for the designation of cells localized in the proliferative zones of the brain. Concurrently, a problem arose concerning the correlation of newly named cells, generally known elements of the nervous tissue, and classic views on its development. In the present study, an attempt is made on the basis of the literature data review and authors’ own experience in the exploration of proliferative zones, to classify cell populations involved in the neurogenesis considering, primarily, the possibilities of their morphological and immunocytochemical in situ identification.
