CHANGES IN Mn-SUPEROXIDE DISMUTASE EXPRESSION IN RAT HIPPOCAMPUS INDUCED BY SINGLE AND TRIPLE EXPOSURE TO MODERATE HYPOBARIC HYPOXIA

Abstract

The purpose of this work was to study the dynamics of expression of mitochondrial Mn-dependent superoxide dismutase (Mn-SOD) 3 and 24 hours after single and triple exposure to mild hypoxia. The investigation was conducted in 18 male Wistar rats using immunocytochemical method. It was shown that in various hippocampal areas the effects of single and triple hypoxia exposure on the Mn-SOD expression could be different or largely similar. The expression dynamics at four time points studied (3 and 24 hours after the first exposure, 3 and 24 hours after the third one) had a wave character which may be important for the development of moderate hypoxia-induced tolerance to subsequent more severe exposures.

FUSION OF FROG CEREBELLAR GRANULE CELLS INDUCED BY TOXIC EFFECTS OF GLUTAMATE AND NO-GENERATING COMPOUND

Abstract

The ultrastructural changes of cerebellar granule cells were studied under stroke modeling conditions, after the toxic effects of glutamate (Glu) and NO-generating compound. Glu toxic doses were shown to induce two types of nuclear chromatin changes. In some cases, the appearance of practically completely decondensed nuclear chromatin was detected, while in the others the nuclei contained partially decondensed chromatin. Pathological fusion of granule cells was observed in both cases. The toxic effect of NO-generating compound on granule cells also caused the appearance of the cells with both completely and partially decondensed (flocculent) chromatin. Granule cells with different chromatin type were able to fuse with each other. Thus, Glu and NO, causing changes in nuclear chromatin, activate the processes of cell clustering with the following cytoplasmic fusion and the formation of multinuclear conglomerates. The possible physiological role of granule cells fusion induced by high concentrations of Glu and NO-generating compound is discussed. This process is considered as a realization of the compensatoryadaptive reactions under extreme conditions observed in the stroke and oxidative stress.

MORPHOLOGICAL CHARACTERISTIC OF GASTRIC SUBMUCOSAL VESSELS IN THE PERITUMORAL AREAS

Abstract

The structural peculiarities of the gastric submucosal vessels in the areas adjacent to the tumor, were studied in 63 patients with gastric cancer. It was found that the processes of tumoral angiogenesis affected not only a tumoral tissue, but also the submucosal regions adjacent to the tumor was characterized by an abnormal structure of its vascular network, their excessive twisting and pathological dilatation. In samples obtained from 33 out of 63 patients studied, the microcirculatory bed vessels were found to be formed by a single layer of endothelial cells. These vessels were found in 25 out of 28 patients with metastases in regional lymph nodes (89,3%) and only in 8 out of 35 patients without metastases (22,8%). The data obtained can be used for predicting the risk of lymphogenic metastase spread in patients with gastric cancer.

Abstract

The changes in the processes of differentiation, proliferation and apoptosis were studied in the culture of human cortical thymocytes after cell exposure to T-32 and T-38 bioregulator peptides. T-32 and T-38 peptides were shown to enhance the differentiation of immature cortical thymocytes (CD4+CD8+) into T-regulatory cells by increasing their proliferate activity and decreasing the level of apoptosis. Moreover, these peptides were found to stimulate the proliferative and antiapoptotic activity of mature T-regulatory (CD4+CD25+) cells.

COMPARATIVE MORPHOLOGICAL STUDY OF THYROID GLAND IN MAMMALIAN ANIMALS AND MAN

Abstract

The objective of this study was to examine the morphological peculiarities of the thyroid gland in animals of different mammalian orders and in man at both macroscopic and microscopic level. 304 specimens of the thyroid gland obtained from the animals of 13 species and 5 mammalian orders and man, were studied. At the macroscopic level, the changes of lobar shape, presence and structural peculiarities of the isthmus, and its relative size were analyzed. At the microscopic level, the following parameters were determined in comparative aspect: the volume fractions of major structural components, the indices of activity, sclerosis and colloid accumulation, the standardized follicular profile shape and size, follicular epithelium height. With the increase of size of the species studied, a tendency was found for the increase of thyroid lobe width and the appearance of parenchymal isthmus, connecting the lobes. At the microscopic level, the variability of follicular profile shape and size was found to increase from small rodents to artiodactyls and man. According to morphological indices, the activity of the thyroid gland in artiodactyls and man was decreased as compared to that one in the other mammalian species.

C-KIT IS A MARKER OF HUMAN PANCREATIC ENDOCRINOCYTE STEM CELLS

Abstract

The aim of our study was to analyze the expression of one of the markers of progenitor cell of different cell types - CD 117 (C-kit) - in human pancreas during prenatal development. The pancreas of human embryos and fetuses at 4-28 weeks of gestation as well as of infants aged up to 2nd postnatal month, was studied. In histological sections, the immunocytochemical reactions were performed with the antibodies against C-kit, insulin and glucagon. In situ hybridization was used for detection of proinsulin mRNA. First cells expressing C-kit were found in human pancreas at 8.5 weeks of gestation among ductal epithelial cells. At 11.5 weeks of gestation these cells were found to segregate from the ductal epithelium and start to form islets. From 8.5 weeks of gestation C-kit positive cells started to express glucagon and proinsulin mRNA, and after 11.5 weeks they also expressed insulin. Islet C-kit positive cells coexpressing both glucagon and insulin, were also found after the birth. It may be concluded that C-kit positive endocrinocyte progenitor cells are common for pancreatic islet A- and B-cells and they are preserved in he islets after the birth.

LIVER REGENERATION AFTER ITS MECHANICAL INJURY IN RATS RECEIVING BIOLOGICALLY ACTIVE SUBSTANCES «TREPEL» AND «SUVAR»

Abstract

The effect of biologically active substances (BAS) «Trepel» and «Suvar» on liver regeneration 1-30 days after its mechanical injury was studied using histological and morphometric methods in 110 rat pups aged 18 days. The control group comprised 90 animals that received no treatment following liver injury. It was shown that both BAS studied inhibited the inflammatory reaction around the injury focus, suppressed collagenogenesis and activated hepatocyte proliferation. This resulted in the significant substitution of the damaged area by the hepatocytes, that was absent in the control animals. In animals treated with BAS, the numbers of binucleated and mitotically dividing hepatocytes was increased, while the amount of hepatocytes with dystrophic changes was reduced. Authors associate the positive effect of BAS on liver structure regeneration with their growth-promoting activity, resulting in the hypertrophy of different organs, including the thyroid and the adrenal glands, that is accompanied by an excessive production of the respective hormones displaying their physiological actions.

Abstract

This investigation was aimed at the study of age changes of penis connective tissue structures. Tissue fragments of penis were obtained from 20 cadavers of men at the age of 20-38 years in group I, and from 20 cadavers of men at the age of 41-59 years in group II. The criteria for the exclusion of material from the research were arterial hypertension, diabetes mellitus, atherosclerosis of internal iliac arteries, Peyronie's disease, and anomalies of genital organ development. It was shown that in the cavernous body of penis, aging was associated with the increased amount and thickening of collagen and argyrophilic fibers, decreased content and thinning of elastic fibers, and the reduced amount of smooth muscle cells (SMC). The average area of fibroblast and SMC nucleolus was not different in both groups studied. The average area of endotheliocyte nucleolus was equal to 1.9±0.9 μm2 in group II, being lower than that one in group I, in which this index was equal to 2.1±0.9 μm2. No differences in the content of type III and IV collagen were found between the study groups. Age-associated decrease in the average area of endothelial cell nucleolus in the cavernous bodies may reflect the reduction of the activity of these cells and may indicate the development of endothelial dysfunction, which is one of the most important steps in the morphogenesis of age-related male erectile dysfunction.

ELECTRON MICROSCOPIC STUDY OF THE HUMAN YOLK SAC EPITHELIUM IN THE I TRIMESTER OF PREGNANCY

Abstract

The ultrastructural organization of the endodermal epithelium of the human yolk sac was investigated during weeks 6 to 12 of intrauterine development. In the beginning of the period studied, the epithelium had features of the tissue specialized both in the active protein synthesis and in the degradation of organic substances transported by endocytosis. Subcellular organization of the yolk sac epitheliocytes is indicative of their functional homology with both the epithelial cells of the small intestine and hepatocytes. By the end of the I trimester of pregnancy, the involutive changes in the human yolk sac took place. The probable mechanisms controlling the organ tissue remodeling are autolysis and apoptosis. A significant ultrastructural remodeling of the endodermal epithelium within the I trimester of pregnancy reflects the short period of human yolk sac activity and is another corroboration of the fast and asynchronous development of extraembryonic organ tissues.

STEREOMORPHOMETRIC CHARACTERISTICS OF HYOID BONE IN ADULT HUMANS WITH DIFFERENT VARIANTS OF NECK STRUCTURE

Abstract

To demonstrate the individual variability of hyoid bone in 50adult men and 50 women with a different variants of neck structure, the following methods were used: anthropometry, anatomical preparation and a stereomorphometry of 100 hyoid bone samples, the correlation and variation-statistical analysis using thoracic-height (T) and cervical (C) indexes. The results have shown that total degree of squares of values of correlation (R) of C with hyoid bone parameters and anthropometry exceeds similar value of T, which confirmed the necessity for subdivision of the individuals into three groups on the basis of C value: in men R=8.585, in women R=11.277; on the basis of Т value - R=3.062 in men, R=4.484 in women. Among the groups, both in men and women, the sum of the squares of correlations of C with anthropometrical parameters and hyoid bone parameters - the maximum value was detected in the groups with an intermediate variant of the neck structure (3.354 and 7.409), the minimal value - in the group with a long and thin neck (1.666 and 3.717). The variation-statistical data are presented on the 7 hyoid bone parameters, which are most significant for morphologists and surgeons. Using the method of σ-deviations of hyoid bone parameters with the consideration of the variants of neck structure, the zones of variability of the average values of hyoid bone were detected. The hypothesis is put forward, stating that the deviation of the average values by more than 3σ is indicative of the unusual phenomenon and could be associated with pathology.

INDIVIDUAL POROSITY CHARACTERISTICS OF HUMAN TUBULAR BONES

Abstract

The diaphyses of the tubular bones of seven men aged 40-60 years without the bone pathology were studied. Porosity characteristics of the compact substance were measured in the tibia and the humerus bones by the method of centrifugal porometry. This method is based on the filling of the objects studied with the wetting fluid and sequential removal of it by the centrifugation at increasing values of the centrifugal acceleration; as a result, the values of the radii of the pores and the respective distribution function of the pore volume could be obtained. The individual features of the bone porosity in different persons were detected, as well as the similarity of porosity characteristics of the bones belonging to one person. Porosity individual features are demonstrated in the form of the distribution curve of pore size and the quantitative distribution of porosity volume according to the pore radii. These features allow to confidently detect the belonging of bones to one person.

COMPARATIVE TOPOGRAPHIC ANATOMICAL ASSESSMENT OF THE OPERATIVE APPROACHES TO THE BODIES OF LUMBAR VERTEBRAE

Abstract

This investigation was aimed at the topographic anatomical comparison of operative approaches to the bodies of lumbar vertebrae according to A.Yu. Sozon-Yaroshevich quantitative criteria and the qualitative characteristic of traumaticity. Modeling of operative approaches according to P.G. Kornev, W.O. Southwick and R.A. Robinson, and V.D. Chaklin was performed on 50 human corpses of both sexes. Five variants of the position of abdominal aorta and inferior cava vein relative to the bodies of vertebrae are described, together with the variants of the interrelation between the lumbar vertebral segment, abdominal aorta and inferior cava vein, 3 variants of aortal bifurcation level and 3 variants of the level of the inferior cava vein formation. No quantitative differences were detected between the right- and left-sided variants of each approach and no significant interrelations between the values of A.Yu. Sozon-Yaroshevich's quantitative criteria and the constitution form (P>0.05). It is shown that from the point of view of topographic anatomical features, P.G. Kornev approach is least favorable. Concerning the approach to LIII, the best topographic anatomical characteristics belong to W.O. Southwick and R.A. Robinson approach, while V.D. Chaklin approach is preferable for the access to LIV.

IMPROVED METHOD FOR THE DEMONSTRATION OF THE CRYSTALLINE LENS CELLULAR STRUCTURES

Abstract

A new method for the demonstration of the lens epithelial cells and fibers in the eye of the experimental animals (rats) is described. The method proposed is not laborious and is simple in utilization. It is based on the intravital administration of methylene blue solution to the animal with the subsequent supravital crystalline lens staining at 37ºC. The material was then embedded into paraffin, and the sections were stained with hematoxylin-eosin. The sections obtained using the proposed method, give an opportunity for the detailed demonstration of the morphological features of the lens cells.
