AGE-DEPENDENT DIFFERENCES OF LPS-INDUCED BDNF AND iNOS IN RAT SUBSTANTIA NIGRA

Abstract

Objective - to study the intensity of Brain-Derived Neurotrophic Factor (BDNF) and inducible NO-synthase (iNOS) expression by substantia nigra glial cells in rats of different ages in norm and in neuroinflammation. Materials and methods. Experiments were performed on 36 male Wistar rats:18 young animals with the body weight of 250- 300 g and 18 old animals weighing 380-420 g. Using immunohistochemical method, these markers were detected in frozen sections in substantia nigra astro - and microgliocytes 8 weeks after stereotaxic unilateral injection of lipopolysaccharide (LPS). into this region. Results: Number of GFAP- and CD11β-positive cells (astro-and microgliocytes respectively) was significantly increased in the substantia nigra of old animals. 8 weeks after endotoxin administration, age-related differences in LPS-induced production of BDNF and iNOS were found. The injection of LPS intensified the production of BDNF in astrocytes in young but not old animals, while at the same time microglial cells of older animals expressed iNOS much more intensively. Conclusions: The balance of trophic and damaging factors synthesized by glial cells of the substance nigra in response to endotoxin stimulation depends on age. The glial response to endotoxin stimulation in young animals has a protective character, whereas in older animals it increases the risk of oxidative damage of neurons.

SURGICAL ANATOMY OF THE INTERCOSTAL NERVES IN THE AREA OF THE LATERAL MARGIN OF THE RECTUS ABDOMINIS MUSCLE

Abstract

Objective - to study the characteristics of the intercostal nerves in the area of the lateral margin of rectus abdominis muscle. Materials and methods. Unfixed cadavers of 88 individuals both sexes without signs of anterior abdominal wall pathology were studied. Among them 45 % were cadavers of men (average age is 53.8±11.9 years) and 55 % of women (51.9±13.2 years). The number of intercostal nerves in the area of lateral margin of aponeurotic sheath of the rectus abdominis muscle was determined, the variants of penetration of intercostal nerves into rectus abdominis muscle were studied, and the distance from the external edge of the rectus abdominis muscle to the point of nerve penetration into its depth was measured. Results. In autopsy material studied, the number of intercostal nerves in the area of rectus abdominis muscle varied from 5 to 7, most frequently being 6 (in 70 %). The occurrence of 5 intercostal nerves was observed significantly more often in men (67 %), while 7 intercostal nerves were more common in women (64 %), and the 6 intercostal nerves were detected almost equally often in both sexes. Besides, posterior (63 %) and lateral (37 %) variants of penetration of the intercostal nerves in the rectus abdominis muscle were found. Posterior variant was significantly more frequently observed in women (69 %), while lateral variant - in men (70 %). The distance between lateral edge of the rectus abdominis muscle and the point of intercostal nerve penetration varied from 1.0 to 4.5 cm and was 3.2±0.4 cm on average. Maximal values of this parameter were observed in the upper area of the anterior abdominal wall and among women. Conclusion. The new data on the surgical anatomy of intercostal nerves in the area of the lateral margin of the rectus abdominis muscle should be considered during the plastic operations for incisional hernia repair.

MORPHOLOGICAL AND FUNCTIONAL CHARACTERISTICS OF LEFT VENTRICULAR MYOCARDIUM IN HIGHLY TRAINED FOOTBALL AND BASKETBALL PLAYERS

Abstract

Objective - to analyze morphological characteristics of left ventricle (LV) myocardium among practicing high-qualification professional athletes specialized in dynamic sports and to evaluate the risk group of cardiac diseases. Material and methods. Echocardiographic study of morphological and functional parameters of LV myocardium was performed and biochemical markers of myocardial damage were analyzed in 65 football and basketball players of high sports qualification. Results. 26 % of athletes examined demonstrated deviation of LV myocardial structure from reference values for young men. Football players were characterized by more profound LV dilation compared to basketball players, with a trend towards increased LV volumes in the group of football players. In 56 % of the athletes, a significant elevation of myocardial damage marker (creatine kinase MB isoform) above the upper limit of normal values was detected. Conclusions. The changes found in athletes may be explained by LV chronic volume overload. High-qualification footballers as compared to basketball players, are characterized by more profound echocardiographic signs of LV overload.

MORPHO-FUNCTIONAL CHARACTERISTICS OF THE MICROCIRCULATORY BED IN DIFFERENT AREAS OF THE GINGIVA

Abstract

Objective - to study blood microcirculation in different parts of the gingiva and to detect its individual-typological characteristics. Materials and methods. Using the methods of biomicroscopy and laser Doppler flowmetry (LDF), the state of microcirculation of blood in different areas of the gingiva was studied in 40 healthy young men aged 17-22 years. The registration of microcirculation was carried out in 24 points of the frontal and masticatory gingiva of the upper and lower jaws. The depth of blood vessel location was studied using histological methods on gingival samples. Results. Data were obtained on the depth and density of capillaries, structural features of the microvessels, and the intensity of tissue blood flow in different areas of the gingiva. With the increase of the distance from the mucogingival fold to gingival margin, the decrease in the density of the capillary networks was observed. Conclusions. Analysis of microcirculation at symmetrical points of the gingiva on the upper and lower jaws showed a low degree of asymmetry in healthy individuals. The predominant type of microcirculation in the gingiva in healthy subjects was the mesohaemic type (87 % of observations).

THE CHARACTERISTICS OF MICROCIRCULATION DURING LOCAL EXPOSURE TO COLD AND THE EFFECT OF REBREATHING TRAINING

Abstract

Objective - to study the effect of hypoxic-hypercapnic training on the morphometric and functional indices of blood microcirculation in young men. Materials and methods. The study involved 80 young men aged 18-21 years, Caucasoid residents of Magadan and St. Petersburg. Before and after training with isolated space breathing free of carbon dioxide (rebreathing), a local cold test was performed, during which one hand was immersed in water with a temperature of 4-5 C for 5 min. Immediately after the test, microcirculation was examined in the nail fold of another hand using computer capillaroscope. Results. It was shown that the morphological characteristics of various capillary regions did not differ in the populations examined. Capillary network density was higher in the residents of Magadan, both before and after cold exposure. However, the speed of blood flow in arterial, venous and transitional parts of the capillaries were lower than in the residents of St. Petersburg. Short-term cooling of the palm did not alter the morphological characteristics of the microcirculation in the intact hand. Rebreathing training did not affect the microcirculation parameters. Conclusions. Cold test and rebreathing training did not affect the microcirculation indices in the nail fold area of the hand, which indicates the relative independence of the reactions of this group of capillaries from systemic hemodynamics and their special role in providing thermal homeostasis during exposure of the body to cold and hypoxia hypercapnia.

Abstract

Objective - to detect the peculiarities of cellular composition of lymphoid nodules of the tracheal wall and to evaluate the neurological status of male Wistar rats with various prognostic resistance to emotional stress on day 1 after hemorrhagic stroke modeling. Materials and methods. After the determination of a resistance to emotional stress in the open field test and subsequent hemorrhagic stroke modeling the lymphoid nodules of the tracheal wall in 42 male Wistar rats were studied histologically. In addition, the neurological status of animals was assessed according to the scale of Menzies et al. (1992). Results. Under the experimental hemorrhagic stroke the destruction of lymphoid cells in tracheal wall lymphoid nodules was detected, which was especially active in stress-prone rats. Already one day after stroke modeling, the reduction of small and medium lymphocytes number was found in the basal portions of lymphoid nodules of stress-prone rats. On the contrary, the number of small and medium lymphocytes in lymphoid nodules was increased in stress-resistant rats. Neurological deficit was present in both groups of rats, with more points of the Menzies scale recorded in stress-prone rats. Conclusions. The data obtained indicate that on day 1 after hemorrhagic stroke modeling the inhibition of immune homeostasis and of the tracheal wall and neurological deficit were observed in stress-prone rats. In the latter, the neurological deficit was more pronounced than in stress-resistant rats.

CHANGES IN THE STRUCTURE AND FRACTAL DIMENSION OF ERYTHROCYTES EXPOSED TO MAGNETITE NANOPARTICLES (AN ATOMIC FORCE MICROSCOPIC STUDY)

Abstract

Objective - to study the basic morphometric characteristics of erythrocytes and to detect their changes after exposure to magnetite nanoparticles. Material and methods. Red blood cells of 23 healthy donors of both sexes aged 20 to 40 years were examined with the use of atomic force microscopy before and after incubation with magnetite nanoparticles for 60 min. Red blood cell diameter, height and fractal dimensions were measured. Results. Incubation with the magnetite nanoparticles caused structural changes in the red blood cells. The main transformed forms were spherocytes, echinocytes, codocytes, stomatocytes. A small amount of forms that could be hardly differented, had perforation of the membrane. Conclusion. Magnetite nanoparticles in sub-lethal concentration cause a significant increase in the number of irreversibly deformed erythrocyte forms. The most sensitive morphometric criterion for differentiating normocytes from all the transformed forms is erythrocyte fractality.

REACTION OF TESTICULAR MAST CELLS TO STRESS OF DIFFERENT INTENSITY

Abstract

Objective - to study structural changes in mast cells of rat testes of in response to exposure to stressors of different intensity. Material and methods. Testes were studied of 20 adult male Wistar rats, subjected to stressful influence of physical loads of different intensity. Mast cells numbers in the testes, their synthetic and functional activity were measured. At the same time, spermatogenesis activity was evaluated on the basis of several criteria. Results. Loads of moderate intensity had no effect on the morpho-functional state of testicular mast cells and didn’t not lead to any disurbances of spermatogenesis, indicating the appearance of training adaptive response. Prolonged exercises of high intensity lead to a decrease in the number of mast cells, reduction of their synthetic activity. Short-term loads of maximum intensity resulted in the decrease of mast cell functional activity. Conclusions. Loads of high and maximal intensity caused changes in morphological and functional parameters of mast cells in the testes of male rats and impeded normal functioning of the seminiferous tubules, providing an inhibitory effect on their sexual function.

Abstract

Objective - to detect the dynamics of the morphometric parameters of hepatocytes after the fracture of the shin bones and to assess the effect of the amino acid mixture on the recovery processes in the liver. Material and methods. Liver was studied in 66 male CBA mice at the age of 2-3 months, divided into 3 experimental groups (54 mice with shin bones fracture) and 1 control group (12 intact mice). The volume of the mononuclear hepatocytes and their nuclei, nuclear-cytoplasmic index, the content of binucleated hepatocytes and their mitotic activity were measured. Observations were conducted on Days 3, 7 and 28 after bone fracture. During the recovery period, mice of the experimental groups received either a standard diet, protein-poor diet or a diet supplemented by an amino acid mixture (with equal weight ratios of leucine, isoleucine, arginine, methionine). Results. Fracture of shin bones was accompanied by a change in the morphometric parameters of hepatocytes, mediated by the organ response to trauma. Peripheral and central zones of the hepatic lobe served as a source of liver reparative regeneration. The use of the amino acid mixture had a stimulating effect on the development of recovery processes in the liver. Conclusions. The liver responds to the bone fractures with the changes of morpho-functional indices of hepatocytes, which differ between the individual zones of the lobules. Dynamics of regenerative processes in the liver depends on the nature of the diet.

COMPARATIVE CHARACTERISTICS OF MAXILLARY MORPHOMETRIC PARAMETERS IN NORMAL AND OBSTRUCTED ERUPTION OF WISDOM TEETH

Abstract

Objective - to examine and compare maxillary morphometric parameters in normal eruption of wisdom teeth and in their retention. Materials and methods. Skulls of 80 adult people were studied that were divided into 4 groups: I - with erupted third molars; II - without the wisdom teeth germs; III - with unilateral retention of upper third molars and IV - with bilateral retention. Results. It was established that in terms of its morphometric parameters, the maxilla with bilateral retention was maximally close to that with missing germs of wisdom teeth, while the maxilla with unilateral retention was close to that with fully erupted third molars. Significant differences between the investigated groups were found in the length of the alveolar arch, the tuberal distance, the distance between prostion and basion, the distance from the incisor to the greater palatine foramina. Along with this, in difficult eruption of third molars, a decrease in the parameters of the bony palate was shown together with the presence of differences in the structure of the gnathic part of the face associated with a more distal maxillar position in retention. Conclusions. Full and timely eruption of wisdom teeth has a significant effect on the maxillary growth, as well as on the vertical profiling of the face.

EARLY STAGES OF REGENERATION HISTOGENESIS IN PERIOSTAL PART OF BONE CALLUS IN MAN

Abstract

Objective - to study the intercellular interactions during early stages of reparative osteohistogenesis. Materials and methods. The object of the study were fracture hematomas, which were obtained from patients of both sexes aged 15-84 years in the course of reconstructive treatment during the first 12 days after the fracture. Results. The earliest event of bone reparation was the migration to the fracture area of a heterogeneous macrophage population with an active cytokine-synthetic function that resulted in the formation of a local pool of VEGF-producing cells in the hematoma and preceded the appearance of the first thin-walled vessels among the tissue detritus. High mitotic activity of the endothelial cell elements in conjunction with the membrane expression of Flk-1 reflected a close molecular interaction between endothelial cells and cytokine-producing macrophages. The active process of angiogenesis occured in parallel with a rapid increase in the amount of connective tissue. By 12 days, a significant decrease in the total number of vessels was noted, which, together with a decrease in mitotic activity of endotheliocytes, reflected a decrease in angiogenesis activity at the time of formation of the structures of woven bone. Expression of Flk-1 and Flt-4 receptors in osteoblastic cells indicated a cytokine regulation of the processes of bone tissue histogenesis. Conclusion. At the early stages of reparative osteogenesis, VEGF-VEGFR regulatory axis was shown to play a key role in the coordination of intercellular molecular interactions, ensuring a consistent replacement of cell-tissue populations in fracture hematoma.

DETECTION OF MORPHOLOGICAL SIGNS OF MAST CELL DEGRANULATION IN HUMAN CHOROID PLEXUS USING DIFFERENT STAINING METHODS AND IMMUNOHISTOCHEMISTRY

Abstract

Objective - to determine the signs of degranulation of mast cells of the human choroid plexus using toluidine blue staining, histochemical reaction to glycosaminoglycans and sulfaminoglycans by alcian blue and immunohistochemical reaction to mast cell tryptase. Materials and methods. The study was performed on fragments of the choroid plexus of the brain taken from 29 individuals aged 14-73 years. To assess the number and location of mast cells, some of the slides were stained with toluidine blue and alcian blue with nuclear fast red counterstaining. Also, immunohistochemical reaction to the human mast cell tryptase was carried out. Results. The applied methods of staining showed different degree of effectiveness in detecting mast cells and their degranulation. It was found that the best method for visualization of mast cells (both granulated and degranulating) in the choroid plexus was tryptase immunohistochemistry. About 60 % of the mast cells in the choroid plexus in the specimens studied were degranulating cells surrounded by an immunopositive halo showing a pericellular localization of tryptase. Conclusions. Tryptase immunohistochemistry was found to be most effective for demonstration mast cells in the choroid plexus of the human brain in comparison to classical methods of toluidine blue and alcian blue staining. The data obtained indicate that degranulation of mast cells in the human choroid plexus belongs to a type of piecemeal degranulation that permanently affects the functional activity of the plexus and, accordingly, the blood-CSF barrier.
